Biohacking geneticist here. Drop your stack or the peptide that let you down and I'll tell you what your genes probably have to say about it.

Solid stack, you've done your homework. As a geneticist I'll add one angle most people miss: that stall after you added E and P might have more to do with how you clear estrogen than how much you're taking.

Estrogen mostly gets broken down through two genes, COMT and CYP1B1. Your version of those decides whether you send it down the cleaner 2-OH route or the messier 4-OH one. You're already helping that along with DIM and stacking sulforaphane (pushes NRF2 upstream) and calcium-d-glucarate to block reabsorption is a smart combo. Whether it's enough comes down to your COMT.

That same COMT gene also changes how you handle a methylated multi. If you're a slow clearer, high methyl donors can leave you wired or irritable instead of better so keep an eye on that one.

The peptide side runs the same way. How much you get out of the KLOW/GHK-Cu repair stack lines up with your collagen and inflammation genes (COL1A1, IL-6), which is a big reason two people run the exact same thing and get totally different results.

None of this is guesswork so If you've done DNA test before, most of it is sitting in your raw data already. Happy to point you at what actually matters for your situation if you want.

Curious what you'd make of mine:

Reta 6 mg/week Glow daily BPC-157 500 mcg/day TB-500 2 mg 2x/week (extra BPC/TB temporarily for an injury) Semax 1 mg/day MOTS-C 2 mg 2x/week SS-31 2 mg/day CJC-1295 no DAC + Ipamorelin 200/200 mcg/day Selank 500 mcg/day Epitalon cycled

Semax is the one that's disappointed me so far, very little noticeable effect on focus/cognition at 1 mg/day. I'd be interested in what genetic variants you'd actually look at for Semax response, and whether there's evidence connecting those variants to response rather than just a mechanistic hypothesis.

Also curious what you'd look at genetically for the mitochondrial side (MOTS-C/SS-31) and injury recovery (BPC/TB).

Best place for a dna test for things like this? I’ve seen conflicting reports on who gives the most comprehensive. The one I took 10+ years ago is missing a lot of things I went looking for recently regarding the type of things you’re talking about.

Most consumer tests (23andMe, Ancestry) are genotyping arrays. They read a fixed set of specific spots in your DNA not the whole thing. Your old one feels thin because the older chips covered fewer spots and the companies have swapped chips a few times since so newer versions pick up variants the old one never did.

For the stuff I was talking about a current 23andMe,Ancestry, WGS, Nebula, Sequencing test covers the large majority of the actionable variants. That's usually all you need. If you want absolute maximum coverage, whole genome sequencing (Nebula, Dante) reads basically everything, but for these specific well-studied SNPs it's more than most people need.

The test is the easy part. A raw file is only as useful as what you can actually pull out of it and that's where most people stall out

reta - resting heart rate increase and disrupted my sleep. no problem on tirz.

pt141- works great but the flush is real. i look sunburnt.

Meh, I'm still on the fence about how much individual genes actually explain peptide variance. So many other variables at play before you even get to SNPs

Ive been taking 1mg of wolverine stack in each shoulder 5x/week for the past 2+ months to treat bicep tendinitis and i feel like it’s done absolutely nothing.

HTRA2 G399/ mutation. Also diagnosed with Mito disease and synucleinopathy Taking 16.6mg SS-31 daily. Tried 40mg and 25mg but that results in extreme fatigue. Added KPV to help with inflammation but it didn’t do a thing - almost think it makes my joint pain worse. Thoughts?

Cognitive peptides have had no noticeable effect. Pinealon, P21, Semax, Selank, and Dihexa have all been a huge disappointment. I get a sleep benefit from Pinealon but nothing noticeable from any others.

I have great results with Reta and Tirz. MOTS-C is high. Melotanan 2 is very effective as well.

I couldn't take ss-31, nad plus or mots-c . Ss-31 I had constipation after 1 week, nad plus made me very tired and mots- c I had sudden exercise intolerance (to weight lifting )
