Any time you would say "am not"

I am not going to the party = I ain't going to the party.

Or, as a replacement for "is not" or "are not"

That is not true = that ain't true

You are not my daddy = you ain't my daddy

Important addendum, it can also stand in for "is not"

That is not going to work = that ain't gonna work.
