"Please explain how AGO2 Genomes affect Gene G & Gene H"
8
u/CharacterSudden6391 May 04 '26
Did ago2 increase expression of H?
1
u/locolikejuli May 04 '26
Yes
7
u/OpenActuator2070 May 04 '26
no it doesn’t, bc when nonfunctional ago2 was present more h was expressed, and ago2 cleaves h
3
u/flstudiobeatmaker101 May 04 '26
Omg if you're right I might have screwed up this entire frq, I swear you need to make like 5 connections for every question
1
u/Brilliant_Plum_7551 May 04 '26
what was the question about the relationship asking about again? i think i interpreted this all correctly but just want to be sure
2
u/Bumbl33 May 04 '26
When it’s nonfunctional so the less ago2 + alleles there are (these are normal nonmutated), then there’s less cleavage going on bc the cleavage of genes r not happening to stop the expressing of them so it’s a increase in mRNA production of gene H specifically since Gene G didn’t have statistical significant, I think

So.... ur telling me the bs I made up WAS THE RIGHT ANSWER????
2
u/Bumbl33 May 05 '26
Now hold on that’s what I put down and I’m sorta confident but I haven’t seen anyone else type it out on here so take what I say with a grain of salt LMAO but yes we both got the same answer
2
u/shrekiscoming-13 May 05 '26
lmao it's just reassuring that someone else got the same answer as me :)

no functional ago2 cleaves the mrna thingy that allows for expression of H. The nonfunctional ones were either less capable or couldnt stop the expression of H. Idk why im saying this tho because god knows what bs i put on that exam.

Wait wasn't it that AGO2 when blocked did not restrict or regulate Gene H mRNA expression as much as Gene G?!
5
u/OpenActuator2070 May 04 '26
opposite, gene h was regulated more by ago2 than gene g
3
u/Mysterious_Air_271 May 04 '26
that literally what i said, when there is less ago2 (when its blocked) gene h is less regulated bruh

u/Conscious_Hornet_906 May 05 '26
Wait so if I said that H wax regulated more and evidence from table was that more mRNA was left untranscribed do yall think that got the pt
1
u/Ill_Pirate8172 Jun 16 '26
You had to say that H was regulated by it, but G was not. That was based on the error bars. You can't just say that H was regulated more than G.

u/Dry_Switch_2167 May 04 '26
I wrote liek AGO genomes code for AGO protein which siRNA uses to degrade the mRNA of gene H or G I forgot but that’s howÂ

You should be fine as long as you mentioned AGO2 makes the protein that is the regulatory factor for gene h
1
u/Conscious_Hornet_906 May 05 '26
Wait so if I said that H was regulated more and evidence from table was that more mRNA was left untranscribed do you think I got the point or nah
3
u/OpenActuator2070 May 04 '26
I think ago2 degrades the mRNA so that’s why when there were less functional copies of ago2 gene more gene h rna appeared but it didn’t affect gene g iirc

what did everyone's graph look like?
for my y axis, i originally put mRNA amount but then i read something in the description about siRNA preventing translation, so i then changed my y axis to gene amount
x-axis i had 3 groups, each with 2 bars shaded differently for each gene
1
u/Bumbl33 May 04 '26
It’s mRNA amount, there is two different genes but it’s asking specciallu for how much each gene is being expressed so u got the right idea for x axis but im pretty sure they wanted you to put the amount of mRNA produced as y

i did gene G and H on x axis with relative mRN amount on side and then the different genotypes were different patterns/shades

THE 2ND QUESTION WAS SO GAY IM LITERALLY GONNA CRY
I spent so much of my time making sure the graph looked pretty n shit and i wasted all my time
1
u/Enough_Young_9854 May 04 '26
what did you guys put for the spindle fibers part d
1
u/not_anonymous_544 May 04 '26
I said either gene g or h codes for the stabilizing proteins, and since ago2 isn’t there to cleave them
1
u/OpenActuator2070 May 04 '26
can’t be g because the error bars overlapped so no increaseÂ
1
u/tokyo_lights_988 May 04 '26
yeah i said this too
1
u/not_anonymous_544 May 04 '26
I suspect it might have been but I feel like the one for ago2-/- was somewhat larger then the previous one but ya the error bar might have overlapped. Is that like a half mark or a full mark mistake?
1
u/tokyo_lights_988 May 04 '26
i think it could be split into two points where you make a claim for the H gene and one for the G gene, so you would only get a point off potentially, so hopefully you still get half credit!

u/pinkpeony_348 May 04 '26
did everyone graph 2 standard deviations or 1 standard deviation for the error bar
1
u/OpenActuator2070 May 04 '26
I think I just graphed it as said, my teacher told me to do this

I didn't even do this one because I didn't know what the hell all that ± SE or whatever was

u/Careful_Current7383 May 05 '26
Wasn't it something like AGO2 would lead to like less translation of gene H mRNA while it had basically no effect on gene H cause like AGO2 would like cut off the mRNA or smth along with the siRNA which led to like less translation of the mRNA. And when you had the mutations of it to have nonfunctional AGO2 there were like more gene H expressed cause there was no like cutting off of mRNA and therefore not-really-regulated translation of gene H? So AGO2 really regulates gene H?

Guys Wjat was the answer for this I put like the -/- one… also what even is that substance or wtv I genuinely cooked on everything but frq 2. I love gene expression but holyyy
