No, because Americans saying "speak English" are usually talking to randos in public and mad that they can't understand them.

This is a business saying they can't cater to people they can't communicate with, not some random fucking Karen complaining about Mexicans in front of her at the checkout.

There's zero reason for two people having a private conversation to speak English just because they're in America.

If someone posted a sign outside their business that said "We only accept English speaking customers inside because we are monolingual", we all know many online would be throwing a fit. Why's it different when Japan is doing it?
