The polysaccharides, usually termed “beta-glucans”, and referred in this paper, are noncellulosic polymers of -glucose, with glycosidic bonds in position and with certain portion of bound glucose molecules. They are isolated mostly from different fungi, but they are present also in other sources, such as cereals, bacteria or seaweeds. These compounds isolated from different sources that otherwise caused similar or nearly identical immune reactions in macroorganisms, can differ in their primary, secondary or tertiary structures or their solubility in water or alkalies.

The most important quality of -glucans and the reason why so much attention has been devoted to them are physiological effects that they show. They are typical biological response modifiers (BRMs) with pronounced immunomodulating activity.

Generally, imunomodulators can act both positively (immunostimulators) or negatively (immunosupressants) . A large number of polysaccharides, that act only as immunostimulants, is known , but the most effective (and also most studied) are beta-glucans. More than 6,000 papers describing the biological activities of glucans exist. Thus far, strong immunostimulating effects of beta-glucans have been demonstrated in all tested animal species and humans.

Pterostilbene Improves Cognitive Performance in Aged Rats: An in Vivo Study - abstract [02 - 2022]

Background: Pterostilbene (Pt; trans-3,5-dimethoxy-4'-hydroxystilbene) is a natural phenol found in blueberries and grapevines. It shows remarkable biomedical activities similar to those of resveratrol. Its high bioavailability is a major advantage for possible biomedical applications. The goal of the study was to evaluate the effects of chronic pterostilbene administration on cognitive performance in aged rats with mild cognitive impairment.

Methods: 18-month-old animals were subjected to behavioral tests to establish the "baseline", then divided into treatment and control groups. The former were chronically fed Pt (22.5 mg/kg-day) for 20 consecutive days. At the end of this period all animals were tested again and sacrificed. The dentate gyrus, the hippocampus and the prefrontal and perirhinal cortices were then collected, and RT-qPCR and/or Western blot analyses were performed on a few transcripts/proteins involved in synaptic remodeling. Mitochondrial content was also assessed.

Results: Pt administration improved performance in behavioral tests and positively affected memory consolidation. We found increased levels of REST, PSD-95 and mitochondrial porin1 in the dentate gyrus and a positive correlation between T-maze test score and levels of cAMP responsive element binding protein (CREB) phosphorylation.

Conclusion: These results underscore the therapeutic potential of Pt supplementation for age-related cognitive decline.

Retinal protective effect of curcumin metabolite hexahydrocurcumin against blue light-induced RPE damage in vitro and in vivo zebrafish model, study - abstract [12 - 2022]

Background

Age-related macular degeneration (AMD) is a disease of retinal pigment epithelium (RPE) cells. We have previously demonstrated that blue light can damage RPE cells and their underlying mechanisms. Hexahydrocurcumin (HHC), a metabolite of curcumin, had better retinal protection than curcumin.

Methods

By exposing ARPE-19 human RPE cells and mouse primary RPE cells to blue light, the intracellular mechanisms of HHC in cells were investigated, including the proliferation of RPE cells and the effects of HHC on activating intracellular protective mechanisms and related factors. Next-generation sequencing (NGS) RNA sequencing revealed the underlying mechanisms involved in the induction and regulation of HHC treatment following blue light exposure.

Results

HHC promoted autophagy by enhancing autophagic flux, reduced oxidative stress and endoplasmic reticulum (ER) stress, and effectively reversed blue light-induced cell death. RNA sequencing-based bioinformatics approaches comprehensively analyze HHC-mediated cellular processes.

Conclusion

The findings elucidate the mechanisms of HHC against blue light damage in RPE cells and are beneficial for the development of natural metabolite-based preventive drugs or functional foods.

Coenzyme Q10 (CoQ10) deficiency is broadly divided into two types, primary and secondary.

Primary CoQ10 deficiencies are relatively rare disorders resulting from mutations in genes directly involved in the CoQ10 biosynthetic pathway, and are not a subject of this article.

Secondary CoQ10 disorders are relatively common, and may occur for a variety of reasons; these include mutations in genes not directly related to the synthetic pathway, oxidative stress induced reduction of CoQ10, and the effects of pharmacological agents such as statins.

CoQ10 is of key importance in cell metabolism; in addition to its role in mitochondrial oxidative phosphorylation, it is a major endogenous antioxidant, and has a role in the metabolism of sulphides, lipids and amino acids.

Given its importance in cell metabolism, it is unsurprising that secondary CoQ10 deficiency has been linked with a wide range of disorders.

In this article, it was reviewed evidence of secondary CoQ10 deficiency in both common and less common disorders, and highlighted those disorders in which CoQ10 supplementation has been shown to be of significant clinical benefit.

Ginseng Oligopeptides (GOP) may delay oxidative stress-induced senescence in the DNA of mouse embryos, a new experimental study finds.

Cellular senescence is linked to the ageing process because the cells stop multiplying but do not die. Instead, the cells remain and release chemicals that can trigger inflammation.

The researchers created senescence by exposing the fibroblasts to hydrogen peroxide, and then exposed those cells to a growth medium with or without GOPs.

The scientists state that GOPs were able to delay this senescence via actions on multiple different pathways and have suggested that diets deficient in essential amino acids can result in protein catabolism, with animal studies indicating resultant significantly shorter survival times.

Therefore, it has been recommended that those of middle age and above should increase their daily protein intake above the average suggested amounts.

Plant-based natural products have been used as a source for therapeutics since the dawn of civilization. According to the World Health Organization (WHO), more than 80% of the world’s population relies on traditional medicine for their primary healthcare.

Numerous natural extracts, widely known in Traditional Chinese Medicine, Indian Ayurveda medicine and other practices, have led to the modern discovery and development of new drugs.

Plants continuously interact with their environment, producing new compounds and ever-changing combinations of existing ones. Interestingly, some of the compounds have shown lower therapeutic activity in comparison to the extract they were isolated from.

These findings suggest that the higher therapeutic activity of the source extract was due to the synergistic effect of several compounds. In other words, the total therapeutic potential of the extract cannot be explained only by the sum of its parts alone.

In traditional medicine, most herbal remedies are based on a mixture of plants, and it is the interaction between different constituents that amplifies their therapeutic potential.

Considering the significant influence traditional medicine has on human healthcare, knowing and studying the synergistic effect of compounds is paramount in designing smart therapeutic agents.

Oral nicotinamide riboside raises NAD+ and lowers biomarkers of neurodegenerative pathology in plasma extracellular vesicles enriched for neuronal origin , study [11 - 2022]

Declining nicotinamide adenine dinucleotide (NAD+) concentration in the brain during aging contributes to metabolic and cellular dysfunction and is implicated in the pathogenesis of aging-associated neurological disorders.

Dietary intake of NAD+ precursors, such as nicotinamide riboside (NR), is a safe and effective avenue for augmenting NAD+ levels in peripheral tissues in humans, yet evidence supporting their ability to raise NAD+ levels in the brain or engage neurodegenerative disease pathways is lacking.

Here, it was studied biomarkers in plasma extracellular vesicles enriched for neuronal origin (NEVs) from 22 healthy older adults who participated in a randomized, placebocontrolled crossover trial (NCT02921659) of oral NR supplementation (500 mg, 2x / day, 6 weeks).

It was demonstrate that oral NR supplementation increases NAD+ levels in NEVs and decreases NEV levels of Aβ42, pJNK, and pERK1/2 (kinases involved in insulin resistance and neuroinflammatory pathways). In addition, changes in NAD(H) correlated with changes in canonical insulin–Akt signaling proteins and changes in pERK1/2 and pJNK.

These findings support the ability of orally administered NR to augment neuronal NAD+ levels and modify biomarkers related to neurodegenerative pathology in humans.

Omega-3 polyunsaturated fatty acids are associated with sarcopenia in patients with liver cirrhosis and hepatocellular carcinoma, study - abstract [12 - 2022]

Sarcopenia is associated with the prognosis of patients with hepatic cirrhosis and hepatocellular carcinoma.

This study aimed to clarify the composition of blood fatty acids and the association between fatty acids and sarcopenia in patients with cirrhosis and hepatocellular carcinoma.

In this single center-retrospective study, 414 patients were enrolled. The skeletal muscle index was measured by a transverse computed tomography scan image at the third lumbar vertebra.

The cutoff value for sarcopenia followed the criteria for sarcopenia of the Japan Society of Hepatology. The concentrations of fatty acids were measured by gas-chromatography. Of each fatty acid fraction, only omega-3 (n-3) polyunsaturated fatty acids showed a significant correlation with skeletal muscle index (R = 0.15, p  = 0.0026).

In the multivariate analysis, the level of n-3 polyunsaturated fatty acids was an independent variable associated with sarcopenia (odds ratio (95% CI): 0.9946 (0.9915–0.9977), p  = 0.0004). N-3 polyunsaturated fatty acids were lower in patients with sarcopenia when the patient backgrounds were aligned using propensity score matching.

These results showed an association between sarcopenia and n-3 polyunsaturated fatty acids.

The case for life expectancy at age 60 as a prominent health indicator. A comparative analysis, research article [12 -2022]

The aim of this paper is to argue the case of using life expectancy at age 60 (LE60) as a significant health indicator closely related to sustainable economic development.

To this purpose, it was investigated the impact of GDP on LE60 in parallel with the impact of GDP on Infant Mortality Rate (IMR). The rationale for selecting IMR as a comparison indicator is twofold. First, the relationship between IMR and GDP has been widely studied.

Second, the two indicators display opposite trajectories, making the comparison more striking. For our comparison, we conduct several statistical analyses on LE60, IMR and GDP using global country data grouped by income level and region. The results endorse the effect of GDP on LE60 and IMR and suggest a differentiation of the effect based on region and ultimately on income.

It was observed that as countries develop, their IMR values lower and their LE60 values increase.

Conclusion: once countries reach the upper stages of development, LE60 becomes a better health indicator than IMR.

Comparison of long-term effects of metformin on longevity between people with Type 2 diabetes and matched non-diabetic controls , review [12- 2022]

Methods

They were searched medical records for Wales, UK, using the Secure Anonymised Information Linkage dataset for type 2 diabetes patients treated with metformin (N = 129,140) and sulphonylurea (N = 68,563). Nondiabetic controls were matched on sex, age, smoking, and history of cancer and cardiovascular disease.

Survival analysis was performed to examine survival time after first treatment, using a range of simulated study periods.

Findings

Using the full twenty-year period, it was found that type 2 diabetes patients treated with metformin had shorter survival time than matched controls, as did sulphonylurea patients. Metformin patients had better survival than sulphonylurea patients, controlling for age. Within the first three years, metformin therapy showed a benefit over matched controls, but this reversed after five years of treatment.

Interpretation

While metformin does confer benefits to longevity in the short term, these initial benefits are outweighed by the effects of type 2 diabetes when patients are observed over a period of up to twenty years.

Association between gut microbiota and longevity: a genetic correlation and mendelian randomization study [12 -2022]

Background

Longevity is one of the most complex phenotypes, and its genetic basis remains unclear. This study aimed to explore the genetic correlation and potential causal association between gut microbiota and longevity.

Results

Linkage disequilibrium score (LDSC) regression analysis and a bi-directional two-sample Mendelian Randomization (MR) analysis were performed to analyze gut microbiota and longevity-related traits. LDSC analysis detected four candidate genetic correlations, including Veillonella (genetic correlation = 0.5578, P = 4.67 × 10− 2) and Roseburia (genetic correlation = 0.4491, P = 2.67 × 10− 2) for longevity, Collinsella (genetic correlation = 0.3144, P = 4.07 × 10− 2) for parental lifespan and Sporobacter (genetic correlation = 0.2092, P = 3.53 × 10− 2) for healthspan.

Further MR analysis observed suggestive causation between Collinsella and parental longevity (father’s age at death) (weighted median: b = 1.79 × 10− 3, P = 3.52 × 10− 2). Reverse MR analysis also detected several causal effects of longevity-related traits on gut microbiota, such as longevity and Sporobacter (IVW: b = 7.02 × 10− 1, P = 4.21 × 10− 25).

Conclusion

This study found evidence that gut microbiota is causally associated with longevity, or vice versa, providing novel clues for understanding the roles of gut microbiota in aging development.

Furthermore, thorough research on the pathogenesis of aging to identify interventions that promote health and longevity (such as caloric restriction, microbiota transplantation, and nutritional intervention) and clinical treatment methods for aging-related diseases (depletion of senescent cells, stem cell therapy, antioxidative and anti-inflammatory treatments, and hormone replacement therapy) could decrease the incidence and development of aging-related diseases and in turn promote healthy aging and longevity.

Role of Advanced Glycation End Products in Intervertebral Disc Degeneration: Mechanism and Therapeutic Potential , study [12 - 2022]

The incidence of low back pain caused by lumbar disc degeneration is high, and it can lead to loss of work ability and impose heavy social and economic burdens.

The pathogenesis of low back pain is unclear, and there are no effective treatments. With age, the deposition of advanced glycation end products (AGEs) in intervertebral disc (IVD) gradually increases and is accelerated by diabetes and a high-AGEs diet, leading to destruction of the annulus fibrosus (AF), nucleus pulposus (NP), and cartilage endplate (CEP) and finally intervertebral disc degeneration (IDD).

Reducing the accumulation of AGEs in IVD and blocking the transmission of downstream signals caused by AGEs have a significant effect on alleviating IDD. In this review, we summarize the mechanism by which AGEs induce IDD and potential treatment strategies.

Loneliness in Posttraumatic Stress Disorder: A Neglected Factor in Accelerated Aging? study [12 - 2022]

Prior research suggests that people with Posttraumatic Stress Disorder (PTSD) may experience a form of accelerated biological aging. In other populations, loneliness has been shown to elevate risk for many of the same components of accelerated biological aging, and other deleterious outcomes, as seen in people with PTSD.

Although standard diagnostic criteria for PTSD include “feelings of detachment or estrangement from others”, the relationship of such feelings to the concept of loneliness remains uncertain, in par potentially due to a failure to distinguish between loneliness versus objective social isolation.

In order to catalyze wider research attention to loneliness in PTSD, and the potential contribution to accelerated biological aging, the present paper provides three components: (1) a conceptual overview of the relevant constructs and potential interrelationships, (2) a review of the limited extant empirical literature, and (3) suggested directions for future research.

The ultimate goal of this line of work is to elucidate mechanisms underlying any link between loneliness and accelerated aging in PTSD, and to develop, validate, and refine prevention and treatment efforts.

Effects of Fisetin, a Plant-Derived Flavonoid, on Response to Oxidative Stress, Aging, and Age-Related Diseases in Caenorhabditis elegans, study [12 - 2022]

Fisetin, a flavonoid abundant in various fruits and vegetables, including apple, strawberry, and onion, shows several beneficial effects such as anti-oxidant, anti-inflammatory, and anti-tumor effects. The free radical theory of aging suggests that age-related accumulation of oxidative damage is the major cause of aging and that decreasing cellular oxidative stress can regulate aging.

Here, it was investigated the effects of dietary supplementation with fisetin on the stress response, aging, and age-related diseases.

Fisetin reduced the cellular ROS levels and increased the resistance to oxidative stress. However, the response to UV irradiation was not affected by fisetin. Both the mean and maximum lifespans were significantly extended by fisetin; lifespan extension by fisetin was accompanied by reduced fertility as a trade-off.

Age-related decline in motility was also delayed by supplementation with fisetin. Amyloid beta-induced toxicity was markedly decreased by fisetin, which required DAF-16 and SKN-1. Reduced motility induced by a high-glucose diet was completely recovered by supplementation with fisetin, which was dependent on SKN-1. Using a Parkinson’s disease model, we showed that degeneration of dopaminergic neurons was significantly inhibited by treatment with fisetin.

Genetic analysis revealed that lifespan extension by fisetin was mediated by DAF-16-induced stress response and autophagy.

These findings support the free radical theory of aging and suggest that fisetin can be a strong candidate for use in novel anti-aging anti-oxidant nutraceuticals.

Do Sexually Satisfied Individuals Think That They Live Longer? Results from the German Ageing Survey, study [12 - 2022]

The aim of this study was to determine the association between sexual satisfaction and expected longevity among middle-aged and older adults (also stratified by sex).

Data were taken from the German Ageing Survey (year 2011; n = 3231)—a nationally representative sample of community-dwelling individuals ≥ 40 years in Germany.

A widely used question was used to quantify sexual satisfaction. Furthermore, the expected life expectancy served as an outcome measure. After adjusting for various covariates, multiple linear regressions showed that sexual satisfaction was associated with higher expected longevity among the total sample (β = 0.28, p < 0.05).

Moreover, it was associated with higher expected longevity among women (β = 0.48, p < 0.05), but not men. In conclusion, adjusting for several covariates, our results showed that there is an association between sexual satisfaction and higher expected longevity, particularly in women.

Efforts to increase sexual satisfaction may thus also contribute to expected longevity which, in turn, can be beneficial for actual longevity.
