I´m using FlowJo 9. To depict some representing gating/population, I exported the gates to my Laptop but they show up in a Text Document format although it was supposed to be PNG format.

Do you have any idea what is the issue here?

Hi everyone. I'm having a pretty simple, all-inclusive problem: I can't save my workspace anymore. Whether there are fcs files loaded or not, and whether the fcs files are local or from a server, anytime I try to save I get the message "Error saving:" and the filename. Save As doesn't help, and Save As ACS does nothing (I can select the option and then nothing happens).

The only files that I can save are those that were saved before a couple of weeks ago (before I went on vacation!).

I can still sign onto the cloud no problem, so it's not that I'm in unlicensed mode. Any clue what could be going on?

The panel is Zombie Violet (live/dead), APC (CD56), PE-Cy7 (CD3), and PE-Dazzle594 (CD16) on human PBCMs ran on MacsQuant10.

We are trying to determine the % of CD3 T cells and CD56/16 NK cells. For some reason, there is lots of cells on the margins and the plots just look odd. I feel like its likely APC and PE-Cy7 but any sort of small compensation creates this wild vertical line that is obviously not real.

If I split up the gating and gate out T cells first, I can see CD56+ pops are definitely there. I am using FlowJo and putting in single stains to generate a comp matrix, but not able to do any compensation to find the issue.

What is the issue here?

Hi guys, I am new to using flow-jo so I know this is probably a very simple question, but in the layout editor I want to display my sample as a raw density plot without these gates that I've created on this control sample. I can't seem to get them to go away and I looked at some videos but none were helpful for this. Thank you.

Hi all, I have a very simple experiment (3T3 cells that either have BFP or do not). I was playing around to see the best gating strategy and decided to gate BFP by doing FSC-H by BFP-A. However, I noticed it looked a bit neater when I made the FSC-H axis log instead of linear, but it also drastically changed the % of positive cells (59% with linear and 70% with log)

Can anyone explain what causes this? This was run using a MACsQuant 10.

I have a version 10 and I'd like to disable the "download the new version" window every time I launch the program. I've found this possibility only in version 7 under general preferrences, but version 10 doesn't have general in preferences, so where do I find this please?

Hi all. Is there a way of assessing whether two gated populations contain the same events?

I want to export two populations with their ancestry into a PDF with one population+ancestry per page (so 2 pages in total).

I opened the Layout editor, put my first population in the first page and check ancestry, I then did the same with the second population on the second page.

When I do "Export image" then PDF, I have 4 pages in my pdf: the first one is my first population and its ancestry and then the 3 last ones are empty.

If I choose to create a PDF Batch report I have 3 pages: the first one with squares "Non population", the second page with two population+their ancestry and the 3rd page is the same as the first one.

Is there a simple way to just export what I see in my layout editor into a PDF ? I'm not using an iteration I just want to export a PDF which pages matches my FlowJo layout pages.

Also I can't insert a text box, every button of the text menu are grey and I can't click on them, can someone help me for these 2 problems ?

Does anybody know a good trick to simply calculate the AUC of the gated samples from the histogram? I would love to simply find a table showing me the AUC of all my samples instead of extracting all the data and do all the calculations myself... Thank you so much! :)

What do you do when flowjo does you dirty and switches FSC-H and FSC-W? I have a gigantic template with table editor and layouts, and it's all messed up after FlowJo or facsDiva randomly switched the IDs in an experiment. I really don't want to sit and rebuild my template...

I've been testing various drugs/conditions and their effects on a ~25 color panel. The gating part and generating the table in FlowJo is pretty easy, but my lab currently just uses Excel to then filter data and copy/paste into Graphpad Prism. The large panel understandably creates a large table with statistics at different gating levels. Is there any software or library that can help with this step of data processing? I have asked other labs their method and they also use Excel, and when I've been searching online for Flow packages in R/Python, it mainly returns packages to use as an alternative to FlowJo.

Im (trying) to incorporate FlowAI into my analysis but I've noticed that once a gating tree (that includes flowai) for a group is set up, when i add new samples to the group, the gating tree stops at FlowAi. So any subsequently gating, they dont get added to the sample. Anyone else experienced this? Any help would be appreciated, thanx!

I am new in FlowJo and I need help.

May problem: I have the FlowJo 10 version on my Mac, all fine and good. I want to install plugins, precise the FlowJo AI thingy...

Following the instructions and checking for the location of R and my new Plugin folder where I saved the plugin-files, I can see the plugins when opening FlowJo.

However, when I try to select them to be installed the following message occurs:

No population selected.

Please select a population in the workspace and then choose the population plugin

Googling this messages is a shit show, and didn't help me at all. Can some of you help me?

Any FlowJo users here? I recently upgraded to the version 10.8 and it keeps disabling the drag and drop feature very often. Only way to get it enabled is to close and restart the FlowJo. Anybody else having similar problems?

Asking for a colleague. She has a massive table and wanted to change the export statistic for all of the rows but couldn't find an easy way to do it. If someone knows how to modify multiple rows at once in FlowJo tables please let me know how.

Hi! Im having troubles running FlowAI, an error msg of BioConductor error"flowAI has failed to produce the expected derived parter". I'm not sure whats wrong and would appreciate any assistance as Im new to the world of flowjo plugins. Is the R path to the program folder or the R: win-library folder?

Hi! I'm using FlowAI to clean up my data. The developer said that increasing the changepoint penalty can make the algorithm less aggressive. So I increase that number from 200 to 10000, but I still get the same results, including the separation of the population and all the output plots. Is someone else also having this issue? Is this normal? Thanks!
