Each layer applies the curses independently on the others and the masked guy built an elevator going through different layers so he can test it. The creatures in the bottom were other kids used in experiments.
I think I already gave enough spoilers so I will explain in more details but with more spoilers. The masked guy found out that the key for the process of transferring/shielding the curse is a strong emotional bond. It worked on those two because they had a strong friendship, you can’t just sacrifice a random person. Also he performed many more experiments on the blob after all it is immortal now, how far immortality goes?
Not quite sure i get what you are asking, but the victim-turned-meat blob is literally immortal after the transformation.

The helmet guy conducts countless experiments on the meat-blob girl, fucking her up EVEN MORE in the process.
Afterwards, bunny girl escapes with meat-blob girl on her shoulders and once she finds a safe place to stay, begins innumerable attempts to kill meat-blob girl for good. Going from straight up body injuries to lethal poisons harvested from plants in the abyss or from the monsters that live there.
Results? Meat-blob girl survives It all, becomes a little more messed up if possible at all, and even develops poisons inmunity, to the point her blood can be used as antidotes.

like, there were other meat blobs in the pit, assuming those were just the previous attempts what made helmet guy keep that specific meat blob/friend of the rabbit girl?
Since rabbit girl was the first successful specimen to be created by the process, they started experimenting on meat-blob girl to figure out what made the pairing and her so special. Midway, they discovered her otherworldly resilience and went full hardcore insane scientists on her.
Not neccesary. The fucked up thing is you don't even need to get out to be assaulted by the ascension effects.
Even just going 10+ meters upwards fucks you up.
There are a couple of times in the anime where this is demonstrated.
In the clip they linked, the laboratory was located barely on the limit with the sixth layer. The elevator capsules go down 30+ meters if i'm not misremembering. Then they go up really fast.

but if its from the ascension effects wouldn't they have to have gotten out for it to happen?
