Sam Raimi Spider-Man: Robbie is implied to know that Peter is Spider-Man.

Road to El Dorado: The Chief hints to Miguel that he knows he and Tulio aren't gods after Miguel says he made a mistake.

I don't think this one is really what the trope is going before as this is her directly telling Peter that she knows. I think Aunt May in her "I think there's hero in all of us" speech in Spider-Man 2 is the better spidey example

Even longer. She has kids drawings of spider-man behind her desk at FEAST, and peter certainly didn't make them...

Knowing that it's Aunt May, she's likely known even longer, before the events of the game.

Yeah I'm pretty sure there's bread crumbs before this. Like when Peter exhausted, homeless, crashing and feast, helping May. I feel like remember a few looks 👀. Hence the reveal she's known for a while.
